大明六字的梵文读音为Om Mani Padme Hum，是观音大士的名号 。“玛呢”解作如意宝珠，指观世音大士之悲心，令所有愿求都能如意得成；“啤咩”解作莲花，指观世音大士如莲花一般清净不染与其清净智。念诵本咒即特别启请大悲观世音菩萨的加持。六字大明咒是“唵啊吽”三字的扩展，其内涵异常丰富、奥妙无穷、至高无上，蕴藏了宇宙中的大能力、大智慧、大慈悲。此咒即是观世音菩萨的微妙本心，久远劫前，观音菩萨自己就是持此咒而修行成佛的，佛名正法明如来。

嗡：读（ōng 嗡），是藏语系佛教的读法。汉语系佛教读“唵”。ōm 是正确读音，OM=AUM，最后要闭唇。
呢：nī 是卷舌音，发音时舌尖要上翘顶上腭，然后放开发?ī音。
叭：bēi
咪：mēi 其它写法有[口+弥]、[口+迷]等。要读成mēi，不能读成 mī，由梵文词的位置格变化而来。 　吽：发音为（hōng轰），也有书注音为（hòng）。发一声还是四声都可以，可根据场景与意愿自行觉定。
